Various diseases or immune pathologies, in particular atopic dermatitis and food allergies, are among manifestations of an imbalance in the composition of gut microorganisms. On top of that, vitamin D is a significant contributor both to modulating the gut microbiome composition and to diverse immune responses from innate and adaptive immunity, which affords ground for studying the relationship between these aspects in the context of the development of atopic conditions. So far, the relationship between the gut microbiota, vitamin D, and the development of these conditions in children has not been sufficiently explored.</p> <p><bold>AIM:<italic> </italic></bold>To study the relationship between the gut microbiota imbalance and immune disorders, in particular the development of food allergies and atopic dermatitis in children; to identify the relationship between bacterial signatures and vitamin D concentrations in children with atopic dermatitis and food allergies.</p> <p><bold>METHODS:<italic> </italic></bold>The 16S ribosomal ribonucleic acid gene of 150 fecal samples from children aged 3 to 12 years was sequenced. The concentration of vitamin D in the participants’ blood serum was determined using enzyme-linked immunosorbent assay. Thereafter, the relationship between certain bacterial signatures and serum 25(OH)D concentration was characterized through the use of statistical models (regression tree, correlation analysis, and linear regression).</p> <p><bold>RESULTS:<italic> </italic></bold>In all mathematical models used, vitamin D concentration turned out to be associated with bacterial families important for atopy, such as <italic>Lachnospiraceae</italic>, <italic>Ruminococcaceae</italic>, and <italic>Sutterellaceae</italic>. The results showed that dysbiosis coupled with vitamin D deficiency is typical of the atopic profile, which may indicate the influence of these conditions on the immune system imbalance toward the Th2 response and, consequently, on the development of allergic conditions.</p> <p><bold>CONCLUSION:<italic> </italic></bold>Certain bacterial patterns specific to atopy are associated with vitamin D concentration in blood serum. In consequence of the study, we concluded that the gut microbiota and vitamin D may be systematically involved in modulating immune responses. Further research is needed to attain full comprehension of the mechanisms underlying these associations and to determine the most efficient interventions for the prevention and treatment of autoimmune conditions.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Обоснование.</bold> Кишечная микробиота и витамин D могут играть важную роль в развитии атопических реакций (атопического дерматита, аллергического ринита, аллергической астмы и пищевой аллергии) у детей. Одним из проявлений дисбаланса в составе микроорганизмов кишечника являются различные заболевания или иммунные патологии, в частности атопический дерматит и пищевая аллергия. Витамин D также играет важную роль как в модуляции состава микробиома кишечника, так и в различных иммунных процессах со стороны врождённого и адаптивного иммунитета, что даёт основания для изучения взаимосвязи данных аспектов в контексте развития атопических состояний. На текущий момент недостаточно изучена взаимосвязь между кишечной микробиотой, витамином D и развитием данных состояний у детей.</p> <p><bold>Цель.</bold> Изучить взаимосвязь дисбаланса микробиоты кишечника с нарушениями иммунитета, в частности с развитием пищевой аллергии и атопического дерматита у детей; выявить взаимосвязь бактериальных сигнатур и концентрации витамина D у детей с атопическим дерматитом и пищевой аллергией.</p> <p><bold>Методы.</bold> Проведено секвенирование гена 16S рибосомной рибонуклеиновой кислоты 150 образцов кала детей в возрасте от 3 до 12 лет. Методом иммуноферментного анализа определена концентрация витамина D в сыворотке крови участников. Затем с помощью статистических моделей (дерево регрессии, корреляционный анализ и линейная регрессия) охарактеризована взаимосвязь определённых бактериальных сигнатур с концентрацией сывороточного 25(OH)D.</p> <p><bold>Результаты.</bold> Во всех использованных математических моделях с концентрацией витамина D оказались связаны такие важные для атопии бактериальные семейства, как <italic>Lachnospiraceae</italic>,<italic> Ruminococcaceae</italic> и <italic>Sutterellaceae</italic>.<italic> </italic>Результаты показали, что дисбактериоз в совокупности с дефицитом витамина D характерен для атопического профиля. Это может свидетельствовать о влиянии данных состояний на дисбаланс иммунной системы в сторону Th2-ответа и, следовательно, на развитие аллергических состояний.</p> <p><bold>Заключение. </bold>Определённые бактериальные паттерны, характерные для атопии, взаимосвязаны с концентрацией витамина D в сыворотке крови. В результате исследования мы пришли к выводу, что кишечная микробиота и витамин D системным образом могут участвовать в модуляции иммунных реакций.
